stains
by traditional concentration and cytocentrifugation method, culture and polymerase chain reaction (PCR) assay for the direct detection of M. tuberculosis.
@ES Methods :
@EN Acid-fast stains by traditional concentration and cytocentrifugation method, culture and PCR assay from 61 sputum specimens were processed according to the standard laboratory procedure. The specimens were liquefied and decontaminated by
N-acetyl-L
cysteine-NaOH(NAIC-NaOH) method. The specimens for the Acid-fast stains were sterilized by 5% sodium hypochlorite. In the PCR assay, a 123-bp segment within the M. tuberculosis specific insertion sequence IS6110 was amplified.
@ES Results :
@EN Comprehensive evaluation of all records of patients such as all laboratory datas, chest x-ray finding and clinical features revealed that 24(39.3%) of 61 sputum specimens were obtained from confirmed tuberculosis patients. On the basis of
this
conclusion, following statistical parameters were evaluated ; overall sensitivity, specificity, positive predictive value, negative predictive value and efficiency of the tests, and which were 91.7, 100, 100, 94.9 and 96.7% respectively for the
culture
method ; 54.2, 100, 100, 77.1 and 82.0% for Acid fast stain by the traditional concentration method ; 79.1, 97.3, 95.0, 87.8 and 90.2% for Acid-fast stain by the cytocentrufugation ; 91.7, 89.2, 84.6, 94.3 and 90.2% for the PCR assay ;
